After evaluating offers from Iowa State, Kansas, Kentucky, and North Carolina, Vaughn ultimately decided to commit to UNLV. During his single season of college basketball for UNLV, he earned the title of Mountain West Conference Freshman of the Year. In 23 games, he showed his skills by averaging 17.8 points and 4.8 rebounds.
On June 25, 2015, Vaughn’s talent led to him being selected by the Milwaukee Bucks as the 17th overall pick in the 2015 NBA draft. Subsequently, he joined the Bucks for the 2015 NBA Summer League and officially signed his rookie scale contract with the team on July 17. Von made his debut for the Bucks in the season opener against the New York Knicks on October 28, contributing 10 points. Off the bench, however, the team lost 122–97.
In a move that shows his versatility, Vaughn signed with Lavrio of the Greek Basket League and Basketball Champions League on January 28, 2022. During his time there, he put up impressive statistics, averaging 14.4 points, 4.6 rebounds and 1 assist. 25 minutes per game in 12 league games. Subsequently, on October 24, 2022, Vaughn joined the training camp roster of the Cleveland Charge. Eventually, on November 20, 2023, he returned to Greece to play for Apollon Patras.
